Dr. Golden's primary clinical activities include treating patients with primary and secondary central nervous system malignancies. In addition, Dr. Golden initiates and enrolls patients with metastatic disease onto immunotherapy and radiation studies. He also initiates and enrolls patients with primary and secondary central nervous system malignancies onto their respective clinical studies.

Dr. Golden is very much involved in translational research. He and his colleagues are pioneers in the use of radiotherapy to individually vaccinate patients against their tumors by promoting tumor-cell immunogenic cell death, thereby inciting immune-mediated abscopal responses. He and his colleagues were the first to show that radiation fosters immunogenic cell death in a dose-dependent manner (Golden et al, Oncoimmunology 2014). This led to the development of the patent titled: assays and methods for assessing immunogenic cell death, efficacy of anti-cancer therapy, and identifying potentially efficacious therapies; application number: PCT/US2014/048385. Additionally, he and his colleagues were the first to report complete abscopal responses in patients with metastatic lung cancer, treated with stereotactic body radiotherapy and immune checkpoint blockade (Golden et al, Cancer Immunology Research 2013; Golden et al, ASTRO. San Antonio, TX 2015). Furthermore, he and his colleagues were the first to show, in a clinical trial, an improved overall survival of patients, who achieved abscopal responses to radiotherapy and GM-CSF (Golden et al, Lancet Oncology 2015). This work helped to lay the foundation for a paradigm shift within the field of radiation oncology, whereby radiotherapy is currently being considered to not only be a local therapy but also a systemic immune-modulating agent.

Prior to this, Dr. Golden helped to elucidate a chemotherapeutic strategy to treat glioblastoma by targeting the endoplasmic reticulum and inducing endoplasmic reticulum stress induced cell death. He and his colleagues were able to show the agonist and antagonistic features of green tea polyphenols in combination with endoplasmic reticulum stress inducing agents (Golden et al, Blood 2009; Chen, Wang, and Golden et al, Cancer Letters 2011). Additionally, Dr. Golden and his colleagues discovered a novel class of autophagy inhibitors capable of improving the efficacy of other endoplasmic reticulum stress inducers (Golden et al, Neurosurgery Focus 2015).
