    A systems biology approach to elucidate the biology of immune-associated outcomes in breast cancer

    Description: The I-SPY 2 neoadjuvant breast cancer clinical trial is a personalized, adaptive trial designed to improve outcomes in high-risk breast cancer patients. This study seeks to develop computational and machine learning (ML) models to unravel the complex interactions between intrinsic host germline and tumor somatic mutations, and extrinsic tumor microenvironment features that drive the biology of response or resistance to therapy. Our study will leverage parallel transcriptomic, genomic, and fluorescence imaging data in the I-SPY2 trial to generate genomic and tumor microenvironment response-predictive biomarkers. Our study aims to improve on current qualifying biomarkers – providing better stratification of patients and directing them to best predicted interventions – and that identify possible alternative treatment strategies for non-responders – reducing unnecessary therapeutic burden.

    Description: We have designed a clinical trial for women treated with drugs for early breast cancer before surgery, in which patients whose cancers do not respond to the initial experimental treatment can be switched to receive a different proven therapy that is considered the best for their particular type of cancer. Our goal is to develop a system that continuously improves our ability to provide each woman with the best chance at being cured, while also trying to avoid any unnecessary treatments that cause toxic side effects.
